Влажные препараты следует хранить там, где они могут иметь свободный доступ к солнечному свету. Если их поместить в темный шкаф, они повреждаются по текстуре и приобретают темный и неприятный цвет; это особенно касается связочных и нервных тканей.
Доктор Брунетти из Падуи, который изобрел новый процесс для сохранения определенных видов препаратов и который получил золотую медаль несколько лет назад на Парижской выставке, дает следующие подробности своего метода:
«Процесс включает четыре отдельные операции, а именно: 1, промывка сохраняемого куска; 2, degraissage, или выедание жирового вещества; 3, дубление и 4, десикация.
«1. Чтобы промыть кусок, М. Брунетти пропускает ток чистой воды через кровеносные сосуды и различные выделительные каналы, а затем вымывает воду током спирта.
«2. Для уничтожения жира он следует за спиртом эфиром, который он проталкивает, конечно, через те же кровеносные сосуды и выделительные протоки; эта часть операции длится несколько часов. Эфир проникает в промежутки плоти и растворяет весь жир. Кусок на этой стадии процесса может храниться любое желаемое время, погружая его в эфир, прежде чем приступать к окончательным операциям.
«3. Для процесса дубления М. Брунетти растворяет танин в кипящей дистиллированной воде, а затем, после вымывания эфира из сосудов дистиллированной водой, вливает этот раствор.
«4. Для процесса сушки доктор Брунетти помещает куски в вазу с двойным дном, наполненным кипящей водой, и заменяет предыдущие жидкости теплым сухим воздухом. С помощью резервуара, в котором воздух сжат до двух атмосфер и который сообщается через кран и систему трубок сначала с вазой, содержащей хлорид кальция, затем с другой нагретой, затем с сосудами и выделительными протоками анатомического куска, находящегося в процессе подготовки, он устанавливает газовый ток, который за очень короткое время вытесняет все жидкости. Операция теперь закончена.
«Кусок остается гибким, легким, сохраняет свой размер, свои нормальные отношения, свои твердые элементы, ибо в нем больше нет никаких жидкостей. С ним можно обращаться без страха, и он прослужит бесконечно».
Для сохранения и отверждения тканей для микроскопического исследования — таких как срезы опухолей, желез, мембран, спинного или головного мозга и т. д. — можно использовать жидкость Мюллера, которая готовится следующим образом: бихромат калия, 75 гран; сульфат натрия, 35 гран; растворенные в шести унциях воды. Препарат следует подвесить в этой жидкости до тех пор, пока он не будет достаточно отвержден, чтобы позволить удовлетворительное исследование.
Подготовка костей.
Патологические препараты костей можно подготовить либо кипячением, либо мацерацией. Метод кипячения, однако, следует использовать только там, где текстура препарата твердая и плотная, как в случаях сросшегося перелома и т. д.; и в этих случаях потребуется осторожность, чтобы избежать повреждения препарата слишком долгим воздействием процесса. Мацерация, однако, является лучшим методом для всех случаев и единственным, который следует использовать в случаях, когда кость размягчена кариесом или некрозом.
Препарат, грубо очищенный от мягких тканей, следует бросить в сосуд с водой, которую меняют каждый день, пока она окрашивается кровью. Теперь его следует оставить до тех пор, пока гнилостный процесс не размягчит ткани настолько, что их можно будет легко удалить, после чего, будучи тщательно промытым в содовой воде, его можно высушить и смонтировать. Если погода холодная, будет абсолютно важно, чтобы мацерация проводилась в теплом помещении, иначе процесс будет настолько медленным, что превратит ткани в форму адипоцира, полностью остановив гнилостный процесс и значительно повредив препарат в его красоте и ценности.
В этой связи можно заметить, что кости от субъекта, который был инъецирован хлоридом цинка, никогда не могут быть успешно мацерированы, так как цинк обладает своего рода дубильным эффектом на ткани, что позволяет им сопротивляться гнилостному процессу. Кипячение — единственный метод, с помощью которого такие препараты могут быть очищены.
Там, где кость была должным образом мацерирована и высушена, и особенно если она от молодого субъекта, она окажется белой и совершенно свободной от жира; но когда она взята от старых субъектов или когда подготовлена кипячением, может потребоваться подвергнуть ее процессу отбеливания для удаления жира и улучшения цвета. Воздействием солнца, частым промыванием в хлорной и содовой воде внешний вид препарата может быть значительно улучшен, или, что еще лучше, путем покрытия препарата на несколько дней серным эфиром, который эффективно растворит весь жир, а затем промыванием и воздействием солнца в течение нескольких дней, он останется красиво белым и чистым.
Для постоянного сохранения препарат следует окончательно смонтировать на блоке или доске таким образом, чтобы наилучшим образом обнажить его интересные моменты.
